Diagnostic Biosystems HER2/NEU C-ERBB-2 c-erbB-2 Oncoprotein
This antibody reacts with the c-erbB-2 oncoprotein, a 190 kDa protein. The proto-oncogene for c-erbB-2 islocated on the human chromosome 17, band 21. It has structural similarities to EGFR. Several studies have indicated that c-erbB-2 may be a good indicator of the prognosis of human carcinomas in the breast, ovary, uterus and gastrointestinal tract.

This antibody is specific to Synacthen (aa1-24 of ACTH); does not react with CLIP (aa17-39 of ACTH). POMC (peo-opiomelanocortin or corticotropinlipotropin) is a 267 amino acid polypeptide hormone precursor that goes through extensive, tissue-specific posttranslational processing by convertases. This antibody stains corticotrophs in the adenohypophysis and is useful in the classification of pituitary adenomas.

Cadherin 17 is exclusively expressed in the epithelial cells of embryonic and adult small intestine, colon, liver, and pancreatic ducts. It has been reported to be frequently expressed in adenocarcinomas arising in the gastrointestinal tract and pancreas. Owing to its restricted expression in these groups of tumors, cadherin 17 has proven to be a useful immunohistochemical marker for assisting in distinguishing these neoplasms from other malignancies with which they may be confused

The antibody is specific against gastrin-containing cells. This anti-body reacts with human gastrin I and human gastrin I fragments. No cross-reactivity was observed with rat gastrin, human big gastrin, human gastrin releasing peptide, sulfated cholecystokinin amide, non-sulfated cholecystokinin amide and cholecystokinin.

Cytokeratins comprise a diverse group of intermediate filament proteins (IFPs) that are expressed as pairs in both keratinized and non-keratinized epithelial tissue, where they constitute up to 85% of mature keratinocytes in the vertebrate epidermis. Cytokeratin 7 is expressed in epithelial cells of ovary, lung and breast. It is often used in conjunction with cytokeratin 20 and CDX-2 in distinguishing pulmonary, ovarian and breast carcinomas (CK7+) from most colon carcinomas (CK7-).

O(6)-methylguanine-DNA-methyltransferase (MGMT) is a 22 kDa human DNA repair protein that removes O(6)-alkylguanine DNA adducts. MGMT acts as a suicide acceptor protein repairing the DNA by accepting alkyl groups and consequently inactivating itself. This antibody stains all mantle zone lymphocytes and 50% of germinal center lymphocytes. Basaloid epithelial cells of tonsilsquamous mucosa are also stained positive with this antibody.

PermaRed/AP is a substrate-chromogen system designed to be used for either IHC or ISH when using alkaline phosphatase detection. PermaRed/AP produces a brilliant dark red color. PermaRed/AP is insoluble in organic solvents; therefore, sections can be dehydrated in alcohol, cleared in xylene (or a xylene-substitute), and permanently mounted. This chromogen substrate system may be used for both automation and manual use.

This antibody reacts with the whole H. pylori. The antibody stains the bacteria present on the surface of the epithelium or in the cytoplasm of the epithelial cells. H. pylori is known to cause peptic ulcer and chronic gastritis in humans.
Diagnostic Biosystems MSH2 Mismatch Repair Protein 2, MSH2
Mutations in DNA mismatch repair genes are associated with hereditary nonpolyposis colorectal cancer (HNPCC). Initially, inherited mutations in the MSH2 and MLH1 homologs of the bacterial DNA mismatch repair genes MutS and MutL were found at high frequency in HNPCC and were shown to be associated with microsatellite instability. The demonstration that 10 to 45% of pancreatic, gastric, breast, ovarian and small cell lung cancers also display microsatellite instability has been interpreted to suggest that DNA mismatch repair is not restricted to HNPCC tumors but is a common feature in tumor initiation or progression.

This product is intended for qualitative immunohistochemistry with normal and neoplastic formalin-fixed, paraffin-embedded tissue sections, tobe viewed by light microscopy. Clinical interpretation of staining results should be accompanied by histological studies with proper controls. Patients’ clinical histories and other relevant diagnostic tests should be utilized by a qualified person (s) when evaluating and interpreting results. This kit is designed to label specific primary antibodies immunohistochemically on tissue sections. The immunohistochemical protocol defined in this brochure is just a guideline. Depending upon tissue fixation conditions, the primary antibody employed, and the user's experience, we encourage individual laboratory to optimize its own protocol. These reagents were tested and quality controlled using tissue sections, however, they can also be optimized for cell smears and cytospin preparations. Sufficient reagents are provided to run 50 to 100 tests.

The Periodic Acid Schiff (PAS) Stain Kit is intended for use in histological demonstration of lymphocytes and mucopolysaccharides. The staining pattern of the lymphocytes are helpful in making therapeutic decisions in established cases of lymphocytic leukemia. The PAS reaction in tissue sections is useful for the demonstration of mucopolysaccharides. PAS staining may also be used for the demonstration of fungal organisms in tissue sections.

The Modified Gomori Methenamine-Silver Nitrate Stain (GMS Stain Kit) is intended for use in the histologic visualization of fungi, basement membrane and some opportunistic organisms such as Pneumocystis carinii. Pneumocystis carinii is an opportunistic pathogen that causes severe pulmonary disease in humans, dogs, rats, mice and other vertebrate species with acquired, induced, or inherited immune deficiency syndromes. In addition, this procedure will demonstrate Actinomyces and related species, Nocardia asteroids, and certain encapsulated bacteria.
